Story Mode in Warriors Orochi utilizes the same chapter selection format from Samurai Warriors while dividing the cast into different factions like in Dynasty Warriors. Each faction has a number of 8 main scenarios and 7 side stories with new characters unlocked upon their completion. Note that side story characters can only be recruited by achieving specific tasks. All of them end up facing Orochi at Koshi Castle albeit with their own unique twists and turns.

Story Mode Chronology
While seemingly self-contained, the main scenarios of each faction are chronologically interwoven with one another despite the game not presenting an explicit timeline. Only the final battle at Koshi Castle serves as a diverging point since the game remains ambiguous over which version of it is considered canon. Side stories are treated as one-shot scenarios that have little to no canonical impact with the overarching plot of this title and its sequels.

Character Unlock Requirements
The unlock methods listed below can be done in any difficulty setting. In the case of characters who happen to be third-party units, players must form an alliance with them before fulfilling the steps needed for their recruitment.
Shu
- Jiang Wei - In Shu Chapter 2-X, successfully trigger Jiang Wei's ambush plot by luring two pairs of enemy officers to the main camp without actually attacking them.
- Ma Chao - In Samurai Chapter 2-X, escort every peasant to the escape route and keep Ma Chao alive.
Wei
- Cao Ren - In Samurai Chapter 5-X, rescue the entire Takeda-Uesugi Army within 2 minutes, escort the cavalry to their designated route, and defeat Cao Ren once he starts moving from his position.
- Pang De - In Shu Chapter 3-X, rescue every Hōjō officer before they surrender to the Orochi Army.
- Sima Yi - In Samurai Chapter 6-X, thwart Sima Yi's ambush plan by defeating Yellowbelly and Urutu before they withdraw back to their destination.
- Zhang He - In Wei Chapter 2-X, escort Cao Pi to Zhang He and have him cooperate in defending the main camp.
- Zhen Ji - In Wei Chapter 6-X, defeat all 6 sorcerers and the impostor Cao Pi under 10 minutes or less.
Wu
- Da Qiao - In Wu Chapter 5-X, meet up with Da Qiao first within 5 minutes before the Orochi Army reaches her.
- Lu Xun - In Samurai Chapter 3-X, escort Lu Xun to the main camp and keep him alive. All allied officers must not be defeated for the duration of the battle.
Other
- Da Ji - Clear every stage available including the side story battles.
- Diao Chan - In Wei Chapter 5-X, meet up with Diao Chan first within 6 minutes and keep her alive.
- Dong Zhuo - In Samurai Chapter 4-X, prevent all 3 supply troops from reaching Dong Zhuo.
- Lu Bu - Impress Lu Bu by defeating him in every faction's Chapter 8-X stage without ever backing away from a direct confrontation.
- Orochi - Unlock every character in the game.
- Zuo Ci - Clear every faction's final chapter.
Samurai 1
- Goemon Ishikawa - In Shu Chapter 4-X, destroy all 10 treasure boxes scattered throughout the map before confronting Goemon.
- Keiji Maeda - In Wu Chapter 6-X, summon all allied reinforcements with the war drum and have them reach the enemy's main camp before 10 minutes are left.
- Kunoichi - In Wu Chapter 4-X, escort all peasants including Yoshimoto to their destination safely. Health items and power-ups are handed out for each successful evacuation.
- Masamune Date - In Shu Chapter 5-X, successfully trigger the pincer attack against Masamune by following each given task correctly without rushing too far into enemy territory.
- Nō - In Wu Chapter 3-X, prevent all enemy messengers from escaping the castle. Cannot be done if Nobunaga is one of the player's team members.
Samurai 2
- Kotarō Fūma - In Wu Chapter 2-X, defeat Kotarō after all of the supply troops have been escorted to the main camp.
- Musashi Miyamoto - In Shu Chapter 6-X, open the flood gates by defeating Lu Bu while ensuring the survival of all third-party officers under Musashi.
- Nene - In Wei Chapter 3-X, defeat Nene's clones within 4 minutes of their appearances.
- Tadakatsu Honda - Impress Tadakatsu by performing certain tasks in every faction's Chapter 7-X stage. During Shu Chapter 7-X, defeat Lu Bu within 2 minutes of his arrival and allow every peasant unit to escape the battlefield. Throughout Wei Chapter 7-X, rescue all Honda officers at the southern fortress, defeat Lu Bu within 2 minutes of his arrival, and clear the stage with Tadakatsu and his officers alive. In Wu Chapter 7-X, defeat Tadakatsu within 2 minutes of his second appearance. For Samurai Chapter 7-X, escort the messenger to the escape route within the northeast stronghold while defeating Lu Bu and keeping every allied officer alive, then make sure that allied reinforcements arrive once the messenger reaches their destination.
- Yoshimoto Imagawa - In Wei Chapter 4-X, prevent all lead engineers from entering the village.
